无忧汤对抑郁模型大鼠海马BDNF和TrkB表达的实验研究 摘要: 抑郁症是一种常见的精神障碍,现有的药物治疗存在一定的局限性,因此寻找有效的药物是非常必要的。无忧汤是一种中药方剂,常用于治疗抑郁症,但其作用机制尚不完全明确。本实验研究了无忧汤在抑郁模型大鼠海马BDNF和TrkB表达上的影响,结果表明,无忧汤能够提高大鼠海马BDNF和TrkB的表达,具有一定的抗抑郁作用。 关键词:无忧汤;抑郁症;BDNF;TrkB;海马 Introduction 抑郁症是一种常见的精神障碍,其主要表现为情绪低落、失眠、食欲减退等。目前抗抑郁药物的应用十分普遍,但其疗效不尽如人意,并且存在一定的副作用。因此,寻找有效的治疗抑郁症的药物是非常有必要的。 无忧汤是一种中药方剂,常用于治疗抑郁症,但其具体作用机制尚不明确。Studieshaveshownthatbrain-derivedneurotrophicfactor(BDNF)andtyrosinekinasereceptorB(TrkB)arecloselyrelatedtothepathogenesisofdepression.BDNFandTrkBareimportantmolecularmarkersofsynaptogenesisandneuroplasticity,andareinvolvedinregulatingthegrowthanddevelopmentofneurons.Therefore,thisstudyaimedtoinvestigatetheeffectofWuyoudecoctionontheexpressionofBDNFandTrkBinthehippocampusofdepressedmodelrats,andtoexploreitsmechanismofaction. MaterialsandMethods Experimentalanimals 60maleSDratswererandomlydividedinto6groups:normalgroup,modelgroup,fluoxetinegroup,andlow,medium,andhighdosegroupsofWuyoudecoction.Theratsinthemodelgroupweresubjectedtochronicunpredictablemildstresstoestablishadepressionmodel. Drugadministration Theratsinthelow,medium,andhighdosegroupsofWuyoudecoctionwereintragastricallyadministeredwith2.5g/kg,5g/kgand10g/kgofWuyoudecoctionrespectivelyfor4weeks.Fluoxetinegroupratswereorallyadministeredwith10mg/kgfluoxetine(Sigma,USA)for4weeks.Thenormalgroupandmodelgroupweregivenequaldosesofnormalsaline. Behavioraltests Openfieldandsucrosepreferencetestswereperformedtoevaluatethebehavioralchangesofratsbeforeandafterdrugadministration.Theopen-fieldtestevaluatestheexploratorybehaviorofratswhilethesucrosepreferencetestevaluatestheirinterestinsugarsolution. Westernblot Aftersacrificingtherats,theirhippocampustissuewascollectedforwesternblotanalysis,andtheexpressionofBDNFandTrkBweredetectedusingspecificantibodies. Results BehavioraltestresultsshowedthatthefluoxetineandWuyoudecoctiongroupshadsignificantlyhigherscoresintheopenfieldtestandsucrosepreferencetestcomparedtothemodelgroup.WesternblotanalysisshowedthattheexpressionofBDNFandTr
