丹皮酚结肠靶向胶囊的制备及评价 Title:PreparationandEvaluationofDanpifenColonTargetedCapsules Abstract: TheaimofthisstudywastoprepareandevaluatecolontargetedcapsulesofDanpifen,awell-knowntraditionalChinesemedicinewithpotentialtherapeuticeffectsoncolonicdiseases.TheformulationwasdevelopedtoensurethetargetedreleaseofDanpifeninthecolon,therebyenhancingitstherapeuticefficacyandreducingpotentialsideeffects. Introduction: Danpifen,alsoknownasChineserhubarb,isawidelyusedtraditionalChinesemedicinewithahistoryofmorethan2,000years.Ithasbeenreportedtopossessvarioustherapeuticproperties,includinganti-inflammatory,antioxidant,andanticancereffects.However,itsbioavailabilityandtherapeuticefficacyarelimitedduetoitspoorsolubilityandlowpermeabilityinthegastrointestinaltract.Therefore,developinganeffectivecolon-targeteddeliverysystemforDanpifenisessentialtoimproveitstherapeuticpotential. Methods: 1.Selectionofexcipients:ExcipientssuchasEudragit®S100,pectin,andguargumwereselectedbasedontheirabilitytoprotectDanpifenfromtheacidicenvironmentofthestomachandpromoteitsreleaseinthecolon. 2.Preparationofcolontargetedcapsules:Thecapsuleswerepreparedbycoatingthedrug-loadedpelletswithamixtureofEudragit®S100,pectin,andguargumusingthefluidizedbedcoatingtechnique. 3.Evaluationofcolontargetedcapsules:Thecapsuleswereevaluatedforvariousparametersincludingdrugcontent,particlesize,coatingefficiency,invitrodrugrelease,andresistancetoacidicconditions. ResultsandDiscussion: 1.Characterizationofcolontargetedcapsules:Thepreparedcapsulesshoweduniformparticlesizedistributionandgoodcoatingefficiency.Thedrugcontentwasfoundtobewithintheacceptablerange. 2.Invitroreleasestudies:ThecolontargetedcapsulesshowedadelayedandsustainedreleaseofDanpifeninsimulatedcolonicfluid,indicatingtheirabilitytobypassthestomachandreleasethedruginthecolon. 3.Resistancetoacidicconditions:Thecapsulesdemonstratedresistancetoacidicconditions,ensuringtheirintactnessinthestomachbeforereachingthecolon. Conclusion: ColontargetedcapsulesofDanpifenweresuccessfull
