黄芩苷铜稳定性分析及其对小鼠实验性肝损伤影响的研究 黄芩苷铜稳定性分析及其对小鼠实验性肝损伤影响的研究 摘要:黄芩苷铜是一种潜在的治疗肝损伤的药物,然而其稳定性在实际应用中尚待进一步研究。本文通过稳定性分析和小鼠实验性肝损伤模型,探讨了黄芩苷铜的稳定性及其对肝损伤的影响。结果显示,黄芩苷铜在不同温度和PH值下的稳定性存在差异,但仍能在常温条件下长时间稳定保存。同时,实验结果表明,黄芩苷铜能够显著降低小鼠实验性肝损伤的程度,减轻肝脏组织的炎症反应和细胞损伤。因此,黄芩苷铜具有潜在的应用价值,但应注意在使用中保持其稳定性。 关键词:黄芩苷铜,稳定性分析,小鼠,肝损伤 Introduction Liverinjury,asacommonclinicalsymptom,hasattractedmoreandmoreattentioninrecentyears.Studieshaveshownthatliverinjuryisrelatedtomanyfactors,includingdrugtoxicity,viralinfection,andalcoholabuse,amongothers.Althoughmanydrugscanbeusedtotreatliverinjury,theyoftenhavevarioussideeffects.Therefore,itisnecessarytodevelopnewdrugswithhighefficacyandfewersideeffectstotreatliverinjury.Huangqinglycosidecopper(HQGC)isapotentialdrugforthetreatmentofliverinjury.However,itsstabilityinpracticalapplicationsneedstobefurtherstudied. Materialsandmethods Stabilityanalysis HQGCwaspreparedbymixingsodiumhydroxide(NaOH),coppersulfate(CuSO4),andHuangqinglycoside(HQG).ThestabilityofHQGCwasanalyzedatdifferenttemperatures(25°C,30°C,37°C,45°C,and70°C)anddifferentpHvalues(4.0,6.0,7.0,and8.0)overaperiodof3months.ThecontentofHQGCwasdetectedbyhigh-performanceliquidchromatography(HPLC)atdifferenttimepoints. Animalexperiments FortyfemaleC57BL/6Jmicewererandomlydividedintofourgroups:control,model,low-dose(5mg/kg),andhigh-dose(10mg/kg)groups.Exceptforthecontrolgroup,allothergroupsweresubjectedtoanexperimentalliverinjurymodelinducedbycarbontetrachloride(CCl4)andintraperitonealinjectionoflipopolysaccharides(LPS).Themiceinthelow-doseandhigh-dosegroupsweregivenHQGCintragastricallyatthecorrespondingdosesfor2weeks.Attheendoftheexperiment,bloodsampleswerecollectedforserumbiochemicalanalysis,andlivertissueswerecollectedforhistopathologicalexamination,quantitativereversetranscription-polymerasechainreaction(qRT-PCR),andwesternblotting.
