Introduction Swiftletnests,alsoknownasbird'snests,arehighlyvaluableintraditionalmedicineandculinarypractices,particularlyinSoutheastAsia.Theyaremadeofsalivaandothermaterialsproducedbyswiftlets,agroupofsmall,insect-eatingbirds.Thenestsarecomposedoffibersthatgluetogetherandformasolidstructure,whichisthenharvestedandsoldinavarietyofforms,suchasraw,dry,orprocessed. Thereareseveralspeciesofswiftletsthatproducenests,withthemostcommonlyusedspeciesbeingAerodramusfuciphagus,Aerodramusmaximus,andCollocaliafuciphaga.However,theidentificationofswiftletnestspeciesisdifficult,astheyaremorphologicallysimilarandtheneststhemselvescanbeprocessedinvariousways,makingitachallengetoidentifyauthenticityandpurity. TheuseofDNAbarcodingandreal-timefluorescencequantitativePCR(qPCR)techniquescanprovideapotentialsolutiontothisproblem.DNAbarcodingisamoleculartechniquethatusesashortDNAsequencefromastandardizedregionofthegenometoidentifyandclassifyspecies.Real-timefluorescenceqPCRisaquantitativemethodthatallowsforthedetectionandquantificationofspecificDNAsequencesinasample.
