Introduction Ovariancanceristhefifthmostcommoncauseofcancer-relateddeathsamongwomen.Themajorityofovariancancercasesarenotdetecteduntilthediseasehasreachedanadvancedstage,resultinginpoorprognosisandhighmortalityrates.Althoughfirst-linetreatmentwithsurgeryandchemotherapycanbeeffectiveintheshort-term,manypatientsexperiencediseaserecurrenceandeventuallysuccumbtothedisease. Intermedin(IMD)isanovelvasoregulatorypeptidethathasbeenshowntohavepotentanti-cancerpropertiesinavarietyofcancermodels.IMDisknowntomodulatecellproliferationandapoptosisthroughmultiplesignalingpathways,includingtheextracellularsignal-regulatedkinase(ERK)pathway. TheERKpathwayisacriticalsignalingcascadethatregulatescellgrowth,differentiation,andsurvival.DysregulationoftheERKpathwayhasbeenimplicatedinthedevelopmentandprogressionofnumerouscancers,includingovariancancer.Therefore,understandingtheeffectofIMDonERKactivityinovariancancercellsmayhelpidentifypotentialtherapeutictargetsforthetreatmentofthisdeadlydisease. MaterialsandMethods Cellculture TheovariancancercelllinesSKOV3andOVCAR3wereobtainedfromtheAmericanTypeCultureCollection(Manassas,VA).CellswereculturedinRPMImediumsupplementedwith10%fetalbovineserum(FBS),100U/mlpenicillin,and100μg/mlstreptomycininahumidifiedincubatorat37°Cand5%CO2. Cellviabilityassay CellviabilitywasassessedusingtheCellCountingKit-8(CCK-8)assay.Cellswereseededin96-wellplatesatadensityof5,000cells/wellandincubatedwithvariousconcentrationsofIMD(0,1,10,50,and100ng/ml)for48hours.Afterincubation,10μlofCCK-8solutionwasaddedtoeachwellandtheplateswereincubatedforanadditional2hours.Absorbancewasmeasuredat450nmusingamicroplater
