Introduction: Phosphorusisavitalnutrientforplantgrowth,anditsavailabilityinsoilplaysacrucialroleinagriculturalproductivity.However,themajorityofphosphorusinsoilsispresentincomplex,insolubleforms,suchasphosphates.Therefore,theefficientutilizationofthisnutrientislimitedduetoitslowsolubility.Phosphate-solubilizingbacteria(PSB)aremicroorganismscapableofconvertinginsolublephosphateintosolubleformsthroughtheprocessofsolubilization.PSBplayasignificantroleinenhancingphosphorusavailabilitytoplantsbysecretingorganicacids,enzymes,andothermetabolites.Inthisstudy,wefocusedonthescreeningandcharacterizationofhigh-efficiencyPSBinearthwormvermicompost. Methods: 1.Collectionandpreparationofearthwormvermicompost:Freshvermicompostsampleswerecollectedandbroughttothelaboratory.Thesampleswerethoroughlymixed,stored,andkeptmoistatroomtemperatureuntilfurtherprocessing. 2.IsolationandscreeningofPSB:Thevermicompostsampleswereseriallydiluted,anddilutionswerespreadonselectivemediacontaininginsolublephosphateasthesolesourceofphosphorus.Plateswereincubatedatanappropriatetemperaturewithproperaeration.Afterincubation,coloniesshowingcharacteristicphosphate-solubilizinghalozoneswereselectedandpurifiedthroughsub-culturing. 3.CharacterizationofPSB:TheselectedPSBisolatesweresubjectedtovariousbiochemicaltests,includingphosphatesolubilizationefficiency,pHtolerance,andtolerancetootherabioticstressfactors.Additionally,moleculartechniquessuchas16SrRNAgenesequencingwereemployedfortaxonomicidentification.
