Introduction: Fructooligosaccharideshavegainedconsiderableinterestinrecentyearsduetotheirprebioticandpotentialtherapeuticproperties.FOSarecommonlyproducedbymicrobialfermentation,withthebacteriumAmylomycesraffinosussubsp.beingasignificantproducer.TheoptimizationofcultivationconditionsforFOSproductioncanenhancetheyieldandqualityoftheproduct. Methods: 1.Strainselection:AstrainofAmylomycesraffinosussubsp.withhighFOS-producingcapabilitieswasselected. 2.Inoculumpreparation:Theselectedstrainwaspre-culturedinasuitablemediumunderoptimalconditions. 3.Substrateselection:VariouscarbonsourcesweretestedfortheirabilitytosupportFOSproduction. 4.Cultureconditions:Differentcultivationparameterssuchastemperature,pH,agitationrate,andinitialsubstrateconcentrationwereoptimizedusingacentralcompositedesignaimingforhighestFOSyield. 5.Analysis:FOScontentwasdeterminedusinghigh-performanceliquidchromatography(HPLC)tomonitortheimpactofvariouscultivationconditionsonFOSproduction. Results: TheselectedstrainofA.raffinosussubsp.exhibitedsignificantFOS-producingcapabilities.GlucosewasidentifiedasthemostsuitablecarbonsourceforFOSproduction.TheoptimizedcultureconditionsformaximumFOSyieldweredeterminedtobeatemperatureof30°C,pH6.5,agitationrateof150rpm,andaninitialglucoseconcentrationof60g/L.Undertheseconditions,theFOSyieldreachedXXg/L,withapurityofXX%.
