CXCL2在肝细胞癌中作用及其机制的研究 Title:TheRoleandMechanismsofCXCL2inHepatocellularCarcinoma Abstract: Hepatocellularcarcinoma(HCC)isoneofthemostcommontypesoflivercancerandaccountsforasignificantnumberofcancer-relateddeathsworldwide.UnderstandingthemolecularmechanismsinvolvedinHCCdevelopmentandprogressioniscrucialforthedevelopmentofeffectivetherapeuticstrategies.Chemokine(C-X-Cmotif)ligand2(CXCL2),encodedbytheCXCL2gene,hasbeenidentifiedasapotentialkeyplayerinHCC. Introduction: Hepatocellularcarcinoma(HCC)isthemajorhistologicalsubtypeofprimarylivercancerandischaracterizedbyuncontrolledproliferationandinvasionofhepatocytes.ThecomplexanddynamicinteractionsbetweencancercellsandthetumormicroenvironmentplayacriticalroleinHCCprogression.Chemokines,smallsecretedproteins,havebeenimplicatedinvariouscellularprocesses,includinginflammation,immuneresponse,angiogenesis,andmetastasis.CXCL2,amemberoftheC-X-Cchemokinefamily,hasrecentlyemergedasanimportantchemokineinHCCpathology. RoleofCXCL2inHCC: CXCL2expressionhasbeenconsistentlyfoundtobeupregulatedinHCCtissuescomparedtonormallivertissues.SeveralstudieshaveshownthatincreasedCXCL2expressioncorrelateswithpoorprognosisandadvancedtumorstageinpatientswithHCC.CXCL2promotesHCCcellproliferation,migration,invasion,andangiogenesis,indicatingitspotentialroleintumorgrowthandmetastasis.Moreover,CXCL2hasbeenimplicatedintherecruitmentandactivationofimmunecellswithinthetumormicroenvironment,contributingtoimmuneescapeandHCCprogression. MechanismsofCXCL2inHCC: MultiplesignalingpathwayshavebeenidentifiedtomediatetheeffectsofCXCL2inHCC.Here,wediscusstwomajorpathways:thenuclearfactor-kappaB(NF-κB)pathwayandthemitogen-activatedproteinkinases(MAPK)pathway.CXCL2activatesNF-κBsignaling,leadingtothetranscriptionalupregulationofgenesinvolvedincellproliferation,survival,andinflammation.ThisactivationpromotesHCCprogression.Additionally,theMAPKpathway,includingtheextracellularsignal-regulatedkinases(ERK)andc-JunN-terminalkinases(JNK)pathways,hasbeenimplicatedinCXCL2-induced
