Introduction Livercancer,alsoknownashepatocellularcarcinoma(HCC),isoneofthemostcommonmalignanciesintheworld.Itisthefifthmostcommoncancerandthethirdmostcommoncauseofcancer-relateddeathsglobally[1].Despitesignificantadvancesindiagnosisandtreatment,theprognosisforlivercancerpatientsremainspoor,witha5-yearsurvivalrateoflessthan20%[2].Oneofthereasonsforthepoorprognosisisthepresenceofcancerstemcells(CSCs)inlivercancer.CSCsareasmallpopulationofcellswithinatumorthathasstemcell-likepropertiesandisresponsiblefortumorinitiation,propagation,andresistancetotherapy[3]. CD133,alsoknownasprominin-1,isaglycoproteinthatisexpressedonthesurfaceofvariousstemcells,includinghematopoietic,neural,andepithelialstemcells[4].Inrecentyears,CD133hasbeenidentifiedasamarkerfortheidentificationandisolationofCSCsinseveraltypesofmalignancies,includinglivercancer[5].SeveralstudieshaveshownthatCD133+cellsinlivercancerarehighlytumorigenicandhaveahigherabilitytoself-renewanddifferentiateintodifferentcelltypes[6].Additionally,theexpressionofCD133hasbeenassociatedwithpoorprognosisinlivercancer[7]. Monoclonalantibodiesarehighlyspecificandsensitivetoolsforthedetectionandcharacterizationofproteins.Inthisstudy,wedevelopedamonoclonalantibodyagainstCD133andanalyzedtheexpressionofCD133antigeninlivercancertissue.
