全氟辛烷磺酸盐诱导斑马鱼胚胎ρ53基因点突变的研究 【Abstract】 Perfluorooctanesulfonicacid(PFOS)isasyntheticfluorinatedcompoundwidelyusedinindustrialandconsumerproducts.IthasbeenreportedtoinduceDNAdamageandchromosomeabnormalitiesinvariousorganisms.Inthisstudy,weinvestigatedtheeffectofPFOSexposureonthemutagenesisoftheρ53geneinzebrafishembryos.OurresultsshowedthatPFOSexposurecouldinducepointmutationsintheρ53gene,suggestingapotentialgenotoxicityofPFOS. 【Introduction】 Perfluorooctanesulfonicacid(PFOS)isasyntheticfluorinatedcompoundwidelyusedinindustrialandconsumerproducts,suchasnon-stickcookware,waterprooffabrics,andfirefightingfoam.Itcanentertheenvironmentthroughvariouspathways,includingair,water,andfood.PFOShasbeendetectedinvariousenvironmentalmatrices,suchaswater,soil,andsediments,aswellasinwildlifeandhumanpopulationsworldwide. PFOShasbeenreportedtoinduceDNAdamageandchromosomeabnormalitiesinvariousorganisms,includingzebrafishembryos.PFOSexposurehasbeenshowntoinduceoxidativestressandDNAdamageinzebrafishembryos,asevidencedbyincreasedlevelsofreactiveoxygenspeciesandDNAdouble-strandbreaks.However,theeffectofPFOSexposureonthemutagenesisofspecificgeneshasnotbeenwellcharacterized. Theρ53geneisatumorsuppressorgenethatplaysacriticalroleinDNAdamageresponseandcellcycleregulation.Mutationsintheρ53genehavebeenassociatedwithvarioushumancancers.Inzebrafish,theρ53genehasbeenshowntobeinvolvedinembryonicdevelopmentandtheresponsetogenotoxicstress.Therefore,themutagenesisoftheρ53geneisanimportantendpointforgenotoxicitytesting. Inthisstudy,weinvestigatedtheeffectofPFOSexposureonthemutagenesisoftheρ53geneinzebrafishembryos. 【MaterialsandMethods】 ZebrafishembryoswereexposedtoPFOSatconcentrationsof0,1,5,and10μg/Lfrom2hpost-fertilizationto24hpost-fertilization.ThePFOSstocksolutionwaspreparedindimethylsulfoxide(DMSO)ataconcentrationof10mg/mL.ThefinalconcentrationofDMSOintheexposuremediumwas0.1%.
