Piezo介导的辅助孵化对小鼠胚胎着床和发育影响的研究 Title:TheImpactofPiezo-assistedAssistedHatchingonMouseEmbryoImplantationandDevelopment Abstract: Assistedhatchingtechniqueshavebeenwidelyusedtoimprovethesuccessratesofassistedreproductivetechnologies.Thisstudyinvestigatestheeffectsofpiezo-assistedhatchingonmouseembryoimplantationanddevelopment.AtotalofXXmouseembryoswererandomlyassignedtothecontrolgroup(CG)orthepiezo-assistedgroup(PAG).TheembryosinthePAGweresubjectedtopiezo-assistedhatching,whiletheCGembryosunderwentnaturalhatchingwithoutanyintervention.Theimplantationrate,embryodevelopment,andgeneexpressionprofileswereevaluated.Ourfindingsrevealedthatpiezo-assistedhatchingsignificantlyenhancedmouseembryoimplantationandoveralldevelopment.Moreover,geneexpressionanalysisdemonstratedalteredexpressionpatternsofgenesrelatedtoembryodevelopmentandimplantationinthePAGgroup.Theseresultssuggestthatpiezo-assistedhatchingcanbeaneffectivetechniqueforimprovingassistedreproductiveoutcomesinmice. Introduction: Assistedreproductivetechnologies(ART)haverevolutionizedinfertilitytreatments,allowingmillionsofcouplestofulfilltheirdreamofparenthood.However,thesuccessratesofARTproceduressuchasinvitrofertilization(IVF)andintracytoplasmicsperminjection(ICSI)remainsuboptimal.Oneofthemajorchallengesisthelowimplantationrateofembryosaftertransferintotheuterus.Variousstrategies,includingassistedhatching,havebeendevelopedtoimprovetheimplantationandpregnancyratesinART.Assistedhatchinginvolvesthemechanicalorlaser-inducedthinningofthezonapellucida,athickproteinshellsurroundingtheembryos,tofacilitatetheirescapeduringhatchingandenhancetheimplantationpotential. Piezo-assistedhatchingisanoveltechniquethatutilizespiezoelectricmicro-manipulatorstocreateasmallholeinthezonapellucida.Theprecisecontrolofferedbypiezo-assistedhatchingallowsforminimaldamagetotheembryoswhilecreatinganopeningfortheirescape.Thistechniquehasbeenwidelyusedinhumaninfertilitytreatments,butitsimpactonmouseembryoimplantationanddevelopmentremainsunderstudied.
