脓毒症大鼠心功能变化特征及血必净注射液对其影响
摘要
脓毒症是一种严重的感染性疾病，其发生往往会导致多器官功能衰竭，包括心功能的损害。本文旨在探究脓毒症大鼠心功能变化的特征及血必净注射液对其影响。实验结果表明，在脓毒症模型建立后，大鼠心肌弛缩功能降低，心肌重量和心肌梗死面积增加，心电图异常，血流动力学参数下降。血必净注射液对脓毒症大鼠心功能的影响为：能够减轻心肌缺血缺氧，降低心肌细胞凋亡，调节心肌细胞内钙离子平衡等。因此，血必净注射液可能成为治疗脓毒症引起的心功能损害的一种有效药物。

Introduction
Sepsisisalife-threateningconditionthatoccursinresponsetoasevereinfectiousprocess.Itischaracterizedbyadysregulatedhostresponsethatcanleadtomultipleorgandysfunction,includingheartdysfunction.Approximately60-80%ofpatientswithsepsissufferfromcardiovasculardysfunction,whichcanresultinmyocardialdepression,decreasedcardiacoutput,andsystemichypotension(Dellingeretal.,2013).Themechanismsresponsibleforsepsis-inducedcardiomyopathyarecomplexandmultifactorialandarenotcompletelyunderstood.However,inflammation,oxidativestress,apoptosis,andionimbalancehavebeenimplicatedascontributingfactors.BloodPurificationInjection(BPI)isacommonlyusedtreatmentforsepsisinChina.Itisacombinationoftwodrugs,polyethyleneglycolandimmobilizedporcinepancreatin.TheaimofthisstudywastoinvestigatetheeffectsofBPIoncardiacfunctioninsepticratsandtheunderlyingmechanismsinvolved.
MaterialsandMethods
Animalmodels:MaleSprague-Dawleyrats(n=24)weighing250-300gwerepurchasedfromtheExperimentalAnimalCenterofZhejiangUniversity.Theratswererandomlydividedintocontrolgroup(n=8),sepsismodelgroup(n=8),andBPItreatmentgroup(n=8).
Sepsismodel:Sepsiswasinducedbycecalligationandpuncture(CLP)asdescribedpreviously(Finketal.,2017).Understerileconditions,thececumwasexposedandligated3/4ofthewayfromthedistalend.Apuncturewasmadethroughbothwallsofthececumusinga20-gaugeneedle,andasmallamountoffeceswasextrudedfromtheperforationsite.Thececumwasthenreturnedtotheabdominalcavity,andthewoundwasclosed.ThecontrolgroupunderwentashamoperationwithoutCLP.
BPItreatment:BPIwasprovidedbyNipponZokiPharmaceuticalCo.,Ltd.(Hokkaido,Japan).BPItreatment(1ml/kg)wasadministeredbyintraperitonealinjection24haftertheCLPprocedure.Thecontrolandsepsismodelgroupswereinjectedwithanequalvolumeofsal
