积雪草酸对高糖培养大鼠肾小球系膜细胞氧化应激及细胞外基质分泌的影响 摘要: 糖尿病是一种常见的代谢性疾病,肾脏是其最常受损的器官之一。高糖水平可导致肾小球系膜细胞氧化应激和细胞外基质过度积累,进而损伤肾小球结构和功能。本文旨在研究积雪草酸(SA)对高糖培养大鼠肾小球系膜细胞氧化应激及细胞外基质分泌的影响。以正常培养和高糖培养的大鼠肾小球系膜细胞为研究对象,利用MTT法检测细胞增殖情况,DCFH-DA染色和ELISA法测定氧化应激指标和细胞外基质分泌物含量。结果显示,高糖培养显著降低了系膜细胞的生长活力,增加了ROS水平,促进了MMP-2和TGF-β1的分泌,而SA处理能够逆转这些变化。此外,SA可以调节Nrf2和HO-1路径,抑制ROS的产生和转录因子NF-κB的活性,从而起到抗氧化和抗炎作用。综上,我们的研究表明,积雪草酸能够保护肾小球系膜细胞,可能具有治疗糖尿病肾病损伤的潜力。 关键词:积雪草酸;高糖培养;肾小球系膜细胞;氧化应激;细胞外基质 Introduction: Diabetesmellitusisacommonmetabolicdisease,andthekidneysareoneofthemostcommonlydamagedorgans.Highglucoselevelscanleadtooxidativestressandexcessivedepositionofextracellularmatrixinmesangialcells,whichdamagesthestructureandfunctionofglomeruli.Mesangialcells,whicharelocatedinthecenteroftheglomerulus,playakeyroleinregulatingbloodflowandmaintainingthestructuralintegrityoftheglomerulus.Oxidativestressandextracellularmatrixaccumulationinmesangialcellsareimportantpathologicaleventsinthedevelopmentofdiabeticnephropathy.Therefore,itisnecessarytofindeffectivedrugstoprotectmesangialcellsandpreventtheprogressionofdiabeticnephropathy. SecuridacainappendiculataHassk,alsocalledJi-xuecaoorSnowflakeLeafflower,isatraditionalChineseherbalmedicinethathasbeenusedtotreatkidneydiseasesforcenturies.Ithasbeenreportedtohaveavarietyofpharmacologicalactivities,includinganti-inflammatory,hypoglycemic,lipid-lowering,andanti-oxidanteffects.SecuridacainappendiculataHasskisparticularlyrichinsecuridacaacid(SA),whichisanoleanane-typetriterpenoidcompound.StudieshaveshownthatSAhasaprotectiveeffectonthekidneysinanimalmodelsofdiabeticnephropathy.However,themechanismofitsprotectiveeffectonmesangialcellsisnotclear.
