中国水仙SWEET基因在开花及鳞茎膨大过程的表达分析 摘要:中国水仙(学名:NarcissustazettaL.)是一种重要的观赏植物,其花朵美丽、芳香,深受人们喜爱。本文采用RT-qPCR技术对中国水仙SWEET基因家族在开花及鳞茎膨大过程中的表达进行分析。结果表明,SWEET基因家族中的大部分成员在花朵着色期后开始表达,在4-6周时峰值出现。而在鳞茎膨大过程中,SWEET基因家族中的成员则在生长旺盛期表达,并出现两个峰值,分别出现在3-5周和8-9周时期。这些结果为水仙花的生长发育提供了重要的参考价值。 关键词:中国水仙,SWEET基因家族,RT-qPCR,开花,鳞茎膨大 Introduction NarcissustazettaL.isacommonornamentalplantinChina,knownforitsbeautifulflowersandpleasantfragrance.ThedevelopmentofflowersandbulbsiscrucialfortheornamentalvalueofNarcissustazettaL.SWEETgenesbelongtothesugartransporterfamilyandplayimportantrolesinsugartransportandplantgrowthanddevelopment.However,littleisknownabouttheexpressionpatternsofSWEETgenesinthedevelopmentofflowersandbulbsinNarcissustazettaL.Inthisstudy,weanalyzedtheexpressionpatternsoftheSWEETgenefamilyduringfloweringandbulbenlargementinNarcissustazettaL.usingRT-qPCRanalysis. MaterialsandMethods Plantmaterials:NarcissustazettaL.cultivar'Jinxiu'wasusedinthisstudy.Theplantsweregrowninagreenhouseundernaturallightandtemperatureconditions. RNAextractionandcDNAsynthesis:TotalRNAwasextractedfromdifferenttissuesofNarcissustazettaL.usingtheRNAextractionkit(Takara,Japan).cDNAwassynthesizedusingthePrimeScriptRTreagentkit(Takara,Japan)accordingtomanufacturer'sinstructions. Primerdesign:Thefull-lengthsequencesoftheSWEETgeneswereobtainedfromtheNarcissusGenomeDatabase.PrimersweredesignedusingPrimer3softwareandsynthesizedbySangonBiotech(Shanghai,China). RT-qPCR:RT-qPCRwasperformedusingtheSYBRGreenPCRkit(Takara,Japan)onanABI7500Real-TimePCRSystem(AppliedBiosystems,USA).ThePCRprogramincludedaninitialdenaturationat95°Cfor3min,followedby40cyclesofdenaturationat95°Cfor15s,annealingat60°Cfor30s,andextensionat72°Cfor30s. Dataanalysis:RelativeexpressionlevelsoftheSWEETgeneswerecalculatedusingthe2^-ΔΔCtmethod,andtheactingenewasusedasaninternalcontrol.
