人参皂甙Rg1预处理对ConA诱导肝损伤的保护作用及机制研究 摘要: 目的:研究人参皂甙Rg1的保护作用及机制,探讨其在ConA诱导肝损伤中的应用价值。 方法:使用ConA注射造成小鼠肝损伤模型,然后给予Rg1预处理,通过检测血清生化参数、肝脏组织病理学变化、肝组织超微结构和相关基因表达等手段研究Rg1的保护作用及机制。 结果:与ConA注射模型组相比,Rg1预处理能够降低血清ALT和AST水平,缓解肝脏组织病理学变化,减轻肝细胞坏死和炎症细胞浸润,改善肝组织超微结构。同时,Rg1预处理下可显著提高SIRT1和Nrf2等抗氧化和解毒相关基因的表达水平,同时抑制NF-κB的激活。 结论:人参皂甙Rg1在ConA诱导的肝损伤中表现出很好的保护作用,其机制可能涉及抗氧化和解毒作用的增强,同时通过抑制NF-κB的活化而缓解炎症反应。因此,Rg1可能成为治疗ConA诱导肝损伤的有效药物之一。 关键词:人参皂甙Rg1;ConA诱导肝损伤;抗氧化;NF-κB Introduction: ConcanavalinA(ConA)isapotentTcellmitogenandcancausesevereliverdamagebyinducingTcell-mediatedimmuneresponse.Manystudieshaveshownthatexcessiveinflammation,oxidativestress,andimmunesystem-mediateddamagearecloselyrelatedtoliverdamagecausedbyConAinjection.Therefore,researchingeffectivetherapiesthatcanalleviatetheConA-inducedliverinjuryisofgreatsignificance. GinsenosideRg1isoneofthemajorbioactiveingredientsinginseng.Ithasbeenfoundtopossessvariousbiologicaleffects,suchasanti-inflammatory,anti-oxidative,andimmunomodulatoryactivities.ThisstudywasdesignedtoinvestigatetheprotectiveeffectsofRg1onConA-inducedliverinjuryanditsunderlyingmechanisms. Materialsandmethods: Animals: MaleKunmingmiceweighing18-22gwereobtainedfromtheExperimentalAnimalCenterofCentralSouthUniversity.AllexperimentalproceduresinvolvingtheanimalswereapprovedbytheInstitutionalAnimalCareandUseCommitteeandwereperformedinaccordancewiththeGuidefortheCareandUseofLaboratoryAnimals. Drugadministration: Rg1wasdissolvedinnormalsalineandadministeredintraperitoneally(10mg/kgor20mg/kg)onceadayforthreeconsecutivedayspriortoConAinjection.MiceintheConAgroupreceivedasingleintravenousinjectionofConA(20mg/kg).MiceintheControlgroupreceivedthesamevolumeofnormalsaline. Samplecollection: Micewereanesthetizedandeuthanizedbycervicaldislocationat24hoursaftertheConAinjection.Bloodandlivertissueswerecollectedforsubsequentanalysis. Assessmentofliverfunction: SerumALTandASTlevelsweremeasuredusingacommercialkitaccordingtothemanufacturer'sinstructions. Histopathologicalexamination: Livertissueswerefixedin10
