载脂蛋白E基因多态性对轻度认知功能障碍患者脑灌注的影响的开题报告 Title:TheInfluenceofApolipoproteinEGenePolymorphismonCerebralPerfusioninPatientswithMildCognitiveImpairment Introduction: Mildcognitiveimpairment(MCI)isacognitivedisorderthataffectsmemory,language,attention,anddecision-makingabilities.ItisconsideredaprecursorofAlzheimer'sdisease(AD),whichcanberelatedtomultiplegeneticandenvironmentalfactors.OneofthemoststudiedgeneticfactorsistheapolipoproteinE(APOE)gene.APOEpolymorphismisassociatedwiththeamountanddepositionofamyloidbeta(Aβ)inthebrain,whichisahallmarkofAD.However,theimpactofAPOEpolymorphismoncerebralperfusioninMCIpatientsisnotwellunderstood. Objectives: TheobjectiveofthisstudyistoinvestigatetheassociationbetweenAPOEgenepolymorphismandcerebralperfusioninMCIpatientsusingabrainMRItechnique. Methods: Thisstudyisacross-sectionalobservationalstudythatwillinvolve100participants,ofwhom50willhaveMCIand50willbehealthycontrols.TheMCIpatientswillbediagnosedaccordingtothecriteriaoftheNationalInstituteonAging-Alzheimer'sAssociationworkgroups.Allparticipantswillundergoaclinicalandneuropsychologicalassessment.ThegenetictestingwillbeperformedusinganRT-PCRmethodtoassessAPOEgenotype(3/3,3/4,or4/4).CerebralperfusionwillbeassessedusinganMRItechniquewitharterialspinlabeling(ASL).Theperfusionmapswillbeconstructedforeachparticipant,andthemeanperfusionvalueswillbeobtainedfromtheregionsofinterest(ROIs)inthebrain.TheROIswillincludethefrontallobes,temporallobes,parietallobes,andhippocampus. Results: ThestatisticalanalysiswillbeperformedusingSPSSsoftware.ThemeanperfusionvaluesoftheROIswillbecomparedbetweentheMCIandcontrolgroupsusingt-testsorMann-WhitneyUtests.TheassociationbetweenAPOEgenotypeandcerebralperfusionwillbeevaluatedusinganalysisofvariance(ANOVA)orregressionanalysis,controllingforage,sex,education,andotherpotentialconfounders. Conclusion: ThisstudywillshedlightontheimpactofAPOEgenepolymorphismoncerebralperfusioninMCIpatients.ThefindingsmayhaveimplicationsfortheearlydiagnosisandtreatmentofAD.
