Inthisproject,weaimtosynthesizeTPredfluorescentorganelleandRNAprobesandtesttheirapplicationinhigh-fidelityfluorescentimagingoflivingcells. Objective OurobjectiveistodesignandsynthesizeTPredfluorescentprobesfororganellesandRNAinlivingcells.Wewilltesttheirspecificity,sensitivity,andphoto-stabilityinvitroandinlivingcells.Finally,wewillusetheseprobestovisualizesubcellularstructuresandobserveRNAexpressioninlivingcellswithhigh-resolutionimaging. Methodology ThesynthesisofredfluorescentorganelleandRNAprobeswillinvolveoptimizingthetargetstructures'chemicalpropertiesandconjugatingthefluorophorestotheprobes'specificDNAorRNAsequences.Wewillusechemicalalterationofthechromophorestructuretoensurehighbrightnessandstability.ThespecificityandsensitivityoftheTPprobeswillbetestedonfixedcellsandcomparedtoconventionalfluorescentprobes.Thephoto-stabilityandTPimagingcapabilitywillbetestedinlivingcells,wherewewillusetechniques,suchasconfocalmicroscopyandFRAPassaystoobservetheprobes'fluorescentproperties.
