Background TheDREB1AgeneisamemberoftheDREBtranscriptionfactorfamily,whichplaysacrucialroleinplantresponsestowaterstress.Ithasbeendemonstratedtoimprovedroughttoleranceinmanyplants,includingArabidopsis,rice,andwheat.InArabidopsis,theoverexpressionofDREB1Aenhancestheexpressionofstress-responsivegenesandimprovesdroughttolerance.Therefore,ithasbeenproposedthattheintroductionofDREB1Aintootherplantscouldhaveasimilareffect. Methods SeveralresearchstudieshavebeenconductedonthetransformationofDREB1Aintovariouscropssuchaswheat,soybean,andtobacco,amongothers,toimprovetheirabioticstresstolerance.However,thereislimitedresearchdoneonthetransformationofDREB1Aintopurpleclover. OnestudyaimedtointroducetheDREB1AgeneintopurplecloverusingtheAgrobacterium-mediatedtransformationmethod.TheresearchersusedaplanttransformationvectorcontainingtheDREB1Ageneandthehygromycinresistancegeneasaselectablemarker.Thetransformedplantswereselectedusingthehygromycinresistancegene,andtheexpressionoftheDREB1Agenewasconfirmedusingquantitativereal-timePCR. AnotherstudyusedtissueculturetoregeneratepurplecloverplantsfromsomaticembryosandthenusedAgrobacterium-mediatedtransformationtointroducetheDREB1Agene.
