Objective: TheaimofthisprojectistoanalyzethegeneticdiversityofcultivatedpeanutusingtheMFLPmolecularmarkerstechnique.Thespecificobjectivesareasfollows: 1.ToextractgenomicDNAfrompeanutcultivarscollectedfromdifferentgeographiclocations. 2.ToPCRamplifytheDNAfragmentsusingMFLPprobes. 3.ToseparatetheDNAfragmentsusingelectrophoresis. 4.ToanalyzethediversityoftheDNAfragmentsusingmolecularmarkerstechniques. 5.Tocomparethegeneticdiversityofthecultivarsfromdifferentgeographiclocations. Methodology: 1.Sampling: Atotalof20peanutcultivarswillbecollectedfromdifferentgeographiclocations,includingAfrica,Asia,andtheAmericas.Thesampleswillbeobtainedfromseedbanks,farms,andresearchcentersthatspecializeinpeanutcultivation. 2.DNAExtraction: GenomicDNAwillbeextractedfromthepeanutcultivarsusingtheCTABmethodasdescribedbyDoyleandDoyle(1987).ThequalityandquantityoftheDNAwillbeassessedusinggelelectrophoresisandaspectrophotometer. 3.MFLPAmplification: TheDNAfragmentswillbeamplifiedusingMFLPprobesspecifictothepeanutgenome.TheMFLPprobesequencesandPCRamplificationconditionswillbeobtainedfromliteratureandoptimizedforthisstudy. 4.Electrophoresis: TheamplifiedDNAfragmentswillbeseparatedusingagarosegelelectrophoresis.Thegelwillbestainedwithethidiumbromide,andtheDNAfragmentswillbevisualizedusingUVtransillumination. 5.DataAnalysis: Themoleculardatageneratedfromtheelectrophoresiswillbeanalyzedusingstatisticalsoftware.Thegeneticdiversityofthepeanutcultivarswillbeassessedusingmolecularmarkerssuchas
