空气放电对莲藕采后腐败菌抑菌机理的研究的中期报告 Title:Studyonthemechanismofairdischargeforinhibitingspoilagebacteriainharvestedlotusroots(Mid-termreport) Introduction: Lotusroot(NelumbonuciferaGaertn)isawidelyconsumedvegetableinAsiancountries.However,lotusrootsarehighlyperishableandsusceptibletopost-harvestdecaycausedbyspoilagebacteria.Theuseofchemicalpreservativesisnotonlyexpensiveandineffectivebutalsoraisesconcernsregardingfoodsafetyandenvironmentalpollution.Inrecentyears,non-thermalplasmatechnology,specificallyairdischarge,hasemergedasapromisingalternativetochemicalpreservativesforthecontrolofspoilagebacteria.Theaimofthisstudyistoinvestigatethemechanismofairdischargeininhibitingspoilagebacteriainharvestedlotusroots. Methodology: 1.Samplecollection:Freshlotusrootswerecollectedfromalocalmarketandtransportedtothelaboratory. 2.Samplepreparation:Thelotusrootswerewashedwithtapwater,peeled,andslicedinto1cmthickdiscs. 3.Treatment:Thelotusrootdiscswereexposedtoairdischargeforvarioustimeintervals(5,10,15,and20minutes)usingacoronadischargesystem. 4.Enumerationofspoilagebacteria:Totalaerobicplatecount,psychrotrophicbacteriacount,andlacticacidbacteriacountweredeterminedusingstandardmicrobiologicalmethods. 5.Measurementofcellmembranepermeability:Cellmembranepermeabilitywasmeasuredusingthepropidiumiodide(PI)stainingmethod. 6.Statisticalanalysis:DatawereanalyzedusingANOVAfollowedbyDuncan'smultiplerangetest. Results: 1.Theresultsshowedthatairdischargetreatmentsignificantlyreducedthetotalaerobicplatecount,psychrotrophicbacteriacount,andlacticacidbacteriacountintheharvestedlotusrootsinatime-dependentmanner(p<0.05). 2.Thecellmembranepermeabilityoftheairdischargetreatedlotusrootswassignificantlyhigherthanthatoftheuntreatedlotusroots(p<0.05),indicatingthatairdischargetreatmentcauseddamagetothecellmembraneofthespoilagebacteria. 3.Theresultsalsoshowedthatlongertreatmenttimesweremoreeffectiveinreducingthebacterialcountsandincreasingthecellmembranepermeability.
