水中微囊藻毒素-LR分子印迹传感技术研究的中期报告 摘要: 微囊藻毒素是一类常见的水生毒素之一,其中微囊藻毒素-LR(MC-LR)由于具有较强的毒性和稳定性,成为了其中研究的热点。本文采用分子印迹技术,以MC-LR为模板分子,通过分别选择甲基丙烯酸甲酯和丙烯酰胺为单体,以过氧化苯甲酰(benzoylperoxide,BPO)为引发剂,制备了分别具有甲基丙烯酸甲酯(MMA)和丙烯酰胺(AM)的聚合物对MC-LR进行分子印迹。采用紫外可见分光光度计测定不同浓度的MC-LR溶液的吸收光谱,通过引入印迹聚合物,在溶液中分别测定印迹聚合物/控制聚合物的吸收光谱,计算了吸光度差值,并绘制了印迹聚合物对MC-LR的选择性系数曲线。结果表明,MMA和AM分别制备的印迹聚合物对MC-LR具有不同的选择性系数,且具有良好的再现性和稳定性。 关键词:分子印迹技术;微囊藻毒素-LR;聚合物;选择性系数 Introduction Microcystinsbelongtotheclassofcyanotoxins,whicharetoxicsecondarymetabolitesproducedbycyanobacteria.Thesetoxinshavebeenidentifiedinaquaticenvironmentsthroughouttheworld,andareaconcernduetotheirpotentialimpactonhumanandanimalhealth.Microcystin-LR(MC-LR)isoneofthemostcommonlydetectedandtoxicvariantsofthesecyanotoxins.Therefore,itiscrucialtodevelopmethodsfordetectingandquantifyingMC-LRinenvironmentalsamples. Molecularlyimprintedpolymers(MIPs)aresyntheticmaterialsthatcanselectivelyrecognizetargetmoleculesduetotheir3Dshapeandfunctionalgrouparrangement,similartoanantibody-antigeninteraction.Inrecentyears,MIPshavebeenincreasinglyutilizedforthedetectionofvariousmolecules,includingtoxins.Inthisstudy,weaimedtodevelopamolecularlyimprintedsensorforthedetectionofMC-LRinwatersamplesusingastrategybasedontheprinciplesofmolecularimprinting. MaterialsandMethods PreparationofMIPs MIPsweresynthesizedbybulkpolymerizationusingmethylmethacrylate(MMA)oracrylamide(AM)asthefunctionalmonomers,MC-LRasthetemplatemolecule,andbenzoylperoxide(BPO)astheinitiator.Thepolymerizationsolutionwaspouredintoglasstubesandpolymerizedat60°Cfor24h.TheMIPparticleswerethencrushedandsievedtoobtainparticleswithasizerangeof300-450μm.
