凋亡抑制蛋白Livin在人乳腺癌中作用的研究的中期报告 摘要： 凋亡抑制蛋白Livin在人乳腺癌细胞中具有重要的生物学功能。本研究旨在探讨Livin在人乳腺癌中的作用及其机制。我们使用PCR和Westernblot检测了Livin在不同的乳腺癌细胞中的表达水平，并验证了Livin的高表达与乳腺癌细胞的增殖、侵袭和转移的相关性。此外，我们利用siRNA沉默Livin基因并检测细胞凋亡和增殖相关分子的表达，确认了Livin的抑制作用与乳腺癌细胞的凋亡有关。最后，我们进行了体内实验，结果显示通过靶向Livin表达能够抑制乳腺癌的生长和转移。综合结果表明，Livin在人乳腺癌中具有促进肿瘤细胞存活、减少凋亡的作用，并且可能成为治疗人乳腺癌的新靶点。 关键词：Livin；乳腺癌；生长；转移；凋亡 Abstract: TheapoptosisinhibitoryproteinLivinplaysanimportantbiologicalroleinhumanbreastcancercells.ThisstudyaimstoexploretheroleandmechanismofLivininhumanbreastcancer.WeusedPCRandWesternblottodetecttheexpressionlevelsofLivinindifferentbreastcancercells,andvalidatedthecorrelationbetweenhighexpressionofLivinandtheproliferation,invasion,andmetastasisofbreastcancercells.Inaddition,wesilencedLivingeneusingsiRNAanddetectedtheexpressionofapoptosisandproliferation-relatedmolecules,confirmingtheinhibitoryeffectofLivinonapoptosisinbreastcancercells.Finally,weconductedinvivoexperiments,andtheresultsshowedthattargetingLivinexpressioncaninhibitthegrowthandmetastasisofbreastcancer.TheresultssuggestthatLivinpromotesthesurvivaloftumorcellsandreducesapoptosisinhumanbreastcancer,andmaybecomeanewtargetforthetreatmentofbreastcancer.
