圆果雀稗内生固氮菌分离及特性研究的中期报告 摘要： 本文介绍了对圆果雀稗内生固氮菌的分离、鉴定以及其在氮素利用方面的特性研究。首先通过对圆果雀稗的根部进行分离培养，筛选出10株内生固氮菌。经过形态学、生理生化特性和16SrDNA序列分析鉴定，确认其中3株分别为苏云金黄色菌株、伸张菌株和根瘤菌株。接下来进行了这些菌株的生理生化特性研究，结果表明它们对温度适应性强、对保护剂敏感、对钙的需求量高等特性。最后对这些分离出的内生固氮菌的固氮能力进行了测定，结果表明这些菌株的固氮能力均相对较强，且在不同环境条件下具有一定的适应性。 关键词：圆果雀稗；内生固氮菌；分离；鉴定；特性研究 Abstract: Thispaperintroducestheisolation,identification,andnitrogenutilizationcharacteristicsofendophyticnitrogen-fixingbacteriainChinesesedge(Carexspp.).Firstly,10strainsofendophyticnitrogen-fixingbacteriawereselectedfromtherootsystemofChinesesedge.Throughmorphological,physiologicalandbiochemicalcharacteristicsand16SrDNAsequenceanalysis,threestrainswereidentifiedasSphingomonassp.,Flavobacteriumsp.,andRhizobiumsp.,respectively.Then,thephysiologicalandbiochemicalcharacteristicsofthesestrainswerestudied,andtheresultsshowedthattheywerehighlyadaptabletotemperature,sensitivetoprotectants,andrequiredhighamountsofcalcium.Finally,thenitrogenfixationabilitiesoftheseisolatedendophyticnitrogen-fixingbacteriaweremeasured,andtheresultsshowedthatthesestrainshadrelativelystrongnitrogenfixationabilitiesandhadadaptabilitytodifferentenvironmentalconditions.
