PDTC对暴发性肝衰竭大鼠肝组织NF-κB表达和细胞凋亡的影响的开题报告 Title:EffectsofPDTConNF-κBexpressionandcellapoptosisinlivertissueoffulminanthepaticfailurerats Introduction: Fulminanthepaticfailure(FHF)isaseriousliverdiseasecharacterizedbyrapidliverfailurewithinashortperiodoftime.FHFisoftenaccompaniedbyhighmorbidityandmortality,therefore,itisurgenttosearchforeffectivetreatmentstrategiesforFHF.PDTC(Pyrrolidinedithiocarbamate)isaselectiveinhibitorofNF-κB,whichisinvolvedinmanyinflammatoryandapoptoticresponses.However,theeffectsofPDTConNF-κBexpressionandcellapoptosisinFHFarestilllargelyunknown.Therefore,thisstudyaimstoinvestigatetheeffectsofPDTConNF-κBexpressionandcellapoptosisinlivertissueofFHFrats. Methods: FHFratswereestablishedbyD-galactosamine(D-GalN)andlipopolysaccharide(LPS)injection.Theratswererandomlydividedintothreegroups:FHFgroup,PDTCgroup(FHFratstreatedwithPDTC)andcontrolgroup(normalrats).Aftertreatment,hepaticfunction,histopathology,cellapoptosisandNF-κBexpressioninthelivertissueweredetected. Results: Comparedwiththecontrolgroup,FHFgroupshowedsignificantliverfunctiondamageandhistopathologicchanges.However,afterPDTCtreatment,theliverfunctionwassignificantlyimprovedandthehistopathologicchangeswererelieved.ThelevelofapoptosisinlivertissuewassignificantlydecreasedinthePDTCgroupcomparedwiththeFHFgroup.Moreover,theexpressionofNF-κBinlivertissuewassignificantlydecreasedinthePDTCgroupcomparedwiththeFHFgroup. Conclusion: PDTCeffectivelyimproveliverfunctionandhistologicpathologyintheFHFrats.PDTCalsoinhibitedNF-κBexpressionandcellapoptosisinlivertissueofFHFrats,suggestingthatPDTCmaybeapotentialtherapeuticagentforFHF.
