Bmp4Smad信号通路在小鼠原始卵泡发育中的作用及作用机制的开题报告 Title:TheRoleandMechanismofBmp4SmadSignalingPathwayinMousePrimordialFollicleDevelopment Introduction: Themammalianovarycontainsafinitenumberofprimordialfolliclesthatserveasthesourceofoocytesforfertility.Thedevelopmentofprimordialfolliclesisacomplexprocessregulatedbyvarioussignalingpathways.Amongthesepathways,thebonemorphogeneticprotein4(Bmp4)signalingpathwayplaysacriticalroleincontrollingfollicularsurvivalandgrowth.Bmp4exertsitseffectsonfolliculardevelopmentbyactivatingtheSmadsignalingpathway.However,theprecisemechanismofBmp4Smadsignalinginprimordialfollicledevelopmentremainsunclear. Objectives: ThisresearchaimstoinvestigatetheroleandmechanismoftheBmp4Smadsignalingpathwayinmouseprimordialfollicledevelopment.Specifically,thisstudyaimstodeterminetheeffectofBmp4onprimordialfolliclesurvival,growth,anddifferentiation,aswellasthedownstreamtargetsoftheSmadpathwaythatmediatetheseeffects. Methods: ThestudywilluseamousemodeltoinvestigatetheroleofBmp4Smadsignalinginprimordialfollicledevelopment.TheovariesofnewbornmicewillbecollectedandculturedinvitrointhepresenceorabsenceofBmp4.TheeffectsofBmp4onfollicularsurvival,growth,anddifferentiationwillbeassessedbyhistologicalexaminationandimmunofluorescencestaining.ThedownstreamtargetsoftheSmadpathwaywillbeidentifiedbytranscriptomeanalysis. Expectedresults: ItisexpectedthatBmp4willpromoteprimordialfolliclesurvivalandgrowth,andinducetheinitiationoffolliculardifferentiation.ThetranscriptomeanalysisisexpectedtoidentifynoveltargetsoftheSmadpathwaythatmediatetheseeffectsandprovideinsightsintothemechanisticbasisofBmp4Smadsignalinginprimordialfollicledevelopment. Conclusion: Thisstudywillcontributetoourunderstandingofthemechanismsthatregulatemammalianfolliculardevelopmentandmayprovidenewtargetsforinfertilitytreatment.
