Functionalstudies ToinvestigatethefunctionofHSPC129,wehavegeneratedHSPC129knockoutmiceusingCRISPR/Cas9geneediting.PreliminaryresultsindicatethatHSPC129knockoutmiceshowdefectsinhematopoieticstemcell(HSC)function,includingimpairedself-renewal,proliferationanddifferentiation.ThissuggeststhatHSPC129iscriticalforthemaintenanceandfunctionofHSCsinmice. WehavealsooverexpressedHSPC129inprimaryhumanHSCsusinglentivirustransduction.OurdatashowthatHSPC129overexpressionenhancesHSCclonogeniccapacityandenhancestheirabilitytogenerateallbloodcelltypes,indicatingapositiveroleforHSPC129inHSCfunction. BioinformaticsanalysishasidentifiedpotentialbindingpartnersforHSPC129,includingproteinsinvolvedinchromatinremodelingandtranscriptionalregulation.Wearecurrentlyinvestigatingtheseinteractionsusingco-immunoprecipitationandmassspectrometrytechniques. Conclusion ThecloningandfunctionalstudiesoftheHSPC129geneprovidenewinsightsintotheregulationofHSCfunction.ByinvestigatingthefunctionofHSPC129anditsinteractionswithotherproteins,weaimtogainadeeperunderstandingofthemolecularmechanismsunderlyingnormalbloodformationandimmunefunction.
