裸鼠前列腺原位肿瘤模型的建立作者：罗勇张林琳贺大林李翔宁亮侯惠莲【关键词】前列腺癌EstablishmentofprostatecancermodelbyorthotopicinjectionofhumanprostatecancercelllineDu145【Abstract】AIM:Toestablishaprostatecancerorthotopicinjectionmodelinnudemiceandtodiscussitsvalueofapplication.METHODS:Lowerabdominalincisionwasmadein10malenudemice.AfterthebladderandprostatewereexposedDu145（6×106）wasinjectedinventralprostatelobesundertheprostaticcapsulebyaTBsyringewitha29gaugeneedleat50μLpermouse.Theabdominalwallandskinwerethenclosedwith70surgicalsutureandthevitalsignsofthenudemicewereobserved.Whenthemiceweredyingtheyweresacrificed.Afterthemousewaskilledthetissuesofprostatebladderlymphaticnodelungandliverweretakenoutfixedin40g/LformaldehydeembeddedinparaffinstainedbyHEandthenobservedmicroscopically.ThebonemetastasiswasobservedbyXray.RESULTS:Theincidenceofprimarytumorafterintraprostaticinjectionwas60%（6of10）.About14dlaterthereweresmallpalpablenodesinprostateandabout19dlatercachexiaoccurredinnudemice.Afterdissectionitwasshownthatthediameterofprimarytumorsvariedfrom0.8cmto1.5cmandtheenvelopeoftheliverbecamecrimpy.Acutehepatitiscouldbeobservedundermicroscopywhichrepresentedalargescaleofhepatocytedeathliversinusdilatationandhyperemiahepaticlobuleinfiltratedbylymphocyteandmacrophageandinconspicuoushyperplasia.CONCLUSION:Theorthotopicinjectionmodelisanoptimalanimalmodeltostudytheoccurrenceanddevelopmentofprostatecancer.Itmimicsthenaturalsituationofhumanprostatecancerandwillhelptounderstandthemechanismsofandrogenindependenceandosseousmetastasisandtumorhostdeterminantsofprostatespecificantigen(PSA)expression.【Keywords】prostateneoplasms;orthotopicinjection;metastasis;nudemice【摘要】目的：建立一种裸鼠前列腺癌原位模型.方法：在10只裸
