Chinese-GermanJournalofClinicalOncologyApril2011,Vol.10,No.4,P232–P234 DOI10.1007/s10330-011-0744-4 ExpressionandclinicalsignificanceofMN1and PTENgeneinpatientswithacutemyeloidleukemia XueshenYan,FanjunMeng,HongguoZhao,JieYang AffiliatedHospitalofMedicalCollege,QingdaoUniversity,Qingdao266003,China Received:23November2010/Revised:22December2010/Accepted:12January2011 ©HuazhongUniversityofScienceandTechnologyandSpringer-VerlagBerlinHeidelberg2011 AbstractObjective:Theaimofourstudywastoexplorethecorrelationsbetweenboththegenes,evolutionandprognosis ofthediseasebydetectingtheexpressionofMN1(meningioma1)geneandPTEN(phosphataseandtensinhomolog)gene inpatientswithacutemyeloidleukemia(AML).Method:MN1andPTENmRNAweredetectedbyreversetranscription-PCR inmononuclearcellsof38patientswithAMLand13patientswithnormalbonemarrow.Results:PositiveratesofMN1and PTENgeneswere76.3%and60.5%respectivelyinbonemarrowmononuclearcells.TheexpressionlevelofMN1mRNAfor thedenovogroupincreasedcomparingwiththatfornormalcontrolgroup(P<0.05),theexpressionlevelofPTENmRNA fordenovogroupdecreasedobviouslycomparingwiththatfornormalcontrolgroup(P<0.01),MN1mRNAleveldecreased inremissiongroup,whilePTENmRNAlevelincreasedcomparingwiththatindenovogroup(P<0.05).MN1mRNAlevel increasedandPTENmRNAdecreasedinrelapsedgroupcomparingwiththatincontrolgroup,andtheirdifferencewas statisticalsignificance(P<0.05).Thetwogenes’levelshadnegativecorrelationsinacutemyeloidleukemia(r=–0.314,P< 0.05).Conclusion:ThereisclosecorrelationsbetweenexpressionofMN1andPTENgenesandtheprognosisandoccur- renceofacutemyeloidleukemia,andtheirexpressioncanbetakenassignificantindexestoaccessthedenovo,relapseand prognosisofacutemyeloidleukemia.
