齐齐哈尔大学毕业设计(论文)齐齐哈尔大学毕业设计(论文)题目香豆素衍生物荧光分子探针的合成及性能研究学院化学与化学工程学院专业班级学生姓名指导教师成绩2012年6月12日摘要以香豆素-3-甲酸为原料,经氯乙酰化得中间体香豆素-3-甲酰氯,进一步与2-吡啶-8-氨基喹啉反应,得荧光分子探针(SHK-1);香豆素-3-甲酰氯与过量的乙二胺反应合成得到SHK-2;2-吡啶-8-氨基喹啉经氯乙酰化得中间体,进一步与无水哌嗪反应,最后与香豆素-3-甲酰氯合成得SHK-3。上述产物的结构经1H-NMR表征,均为目标产物。经初步筛选后,研究了荧光分子探针SHK-1在中性缓冲溶液中对金属离子的识别性能。结果表明,SHK-1可以选择性荧光猝灭识别Cu2+、Fe3+。SHK-1与Fe3+络合后,SHK-1在波长为351nm处,荧光峰强度猝灭31%;与Cu2+络合后,SHK-1在474nm处,荧光峰强度几乎完全猝灭,猝灭比率达97%。定量分析显示,SHK-1与Cu2+形成了1:1型络合物。金属离子竞争实验表明,常见的金属离子均不干扰SHK-1对Cu2+的检测。关键词:香豆素;荧光分子探针;Cu2+AbstractCoumarin-3-methylchloridewassynthesizedusingtherawmarterialcoumarin-3-formicacidthroughchloroacetylation.whichwereusedasmediumsofsynthesizingcomplicatedfluorescentprobes.Furtherly,usedcoumarin-3-methylchlorideand2-pyridine-8-aminoquinoline,fluorescentmolecularprobesSHK-1wasdesignedandsynthesizedthroughreaction.SHK-2wassynthesizedusingthecoumarin-3-methylchlorideandexcessethylenediamine..2-pyridine-8-aminoquinolinewaschloroacetylatedusingtherawmarterial,whichisthemediumsofsynthesizingcomplicatedfluorescentprobes.Andthenfurtherreactionwithanhydrouspiperazine,finallytheSHK-3wassynthesizedthroughcoumarin-3-methylchloride.Thestructurewasconfirmedby1H-NMRcharacterizationoftheaboveproducts,arethetargetproduct.Afterinitialscreening,fluorescentmolecularprobesoftheSHK-1wasstudiedintherecognitionperformanceofthemetalionsintheneutralbuffersolution.TheresultsshowthatSHK-1canbeselectivefluorescencequenchingidentificationofCu2+,Fe3+.AftercomplexationwithFe3+,atthewavelengthof351nm,thefluorescencepeakintensityquenchingof31%;AftercomplexationwithCu2+,atthewavelengthofthe474nm,Thefluorescenceintensityisalmostcompletequenching,thequenchingratioof97%.QuantitativeanalysisshowedthatSHK-1formeda1:1complexwithCu2+.MetalioncompetitionexperimentsshowthatthedetectionofSHK-1forCu2+didnotbeeninterferedforthecommonmetalions.Keywords:Coumarin;Fluorescentmolecularprobes;Cu2+目录TOC\o"1-3"\h\z\uHYPERLINK\l"_Toc265315235"摘要PAGEREF_Toc265315235\hIHYPERLINK\l"_Toc265315236"Abstract
