褪黑素对PTZ致痫大鼠海马神经元凋亡的影响褪黑素对PTZ致痫大鼠海马神经元凋亡的影响【摘要】 目的 探讨褪黑素对癫痫发作中细胞凋亡的影响。方法 将24只成年Wistar大鼠(180～230g)随机分为NC组、PTZ组和MT组；PTZ组和MT组腹腔注射PTZ(60mg/kg)，诱导癫痫发作，NC组腹腔注射等量生理盐水。MT组按体重10mg/kg于注射PTZ之前20min、立即、后1h、后2h分别经腹腔注射MT并观察1h，PTZ组于相同时间注射生理盐水。然后处死大鼠取脑，应用免疫组织化学的方法检测海马神经元Bax和Bcl-2蛋白的表达。结果 大鼠海马神经元Bax的改变：PTZ组明显高于NC组，MT组明显低于PTZ组，但高于NC组：大鼠海马神经元Bcl-2的改变：PTZ组明显高于NC组，MT组明显高于PTZ组，也高于NC组。结论 MT可以通过增强海马神经元Bcl-2的表达和降低Bax的表达而对抗癫痫中细胞的凋亡。      【关键词】 褪黑素；癫痫；戊四氮；Bax；Bcl-2       【Abstract】 Objective Toexploretheeffectofmelatoninonbcl-2andbaxofhippocampusinseizuresratsinducedbyPTZ.Methods Twenty-fouradultsWistarrats(180～230g)wererandomlypidedintothreegroups，NCgroup，PTZgroupandMTgroup.AdoseofPTZ(60mg/kg)wasintraperitoneallyinjectedintotheratsofPTZgroupandMTgroupandseizureswasinduced.TheratsofNCgroupwereadministeredwiththesamedoseofnormalsaline.TheratsofMTgroupwereadministeredintraperitoneallywithMT(10mg/kg)in20minutebeforeinjectingPTZ，atonetime，1and2hourafterinjection.SimultaneouslythesamedoseofnoramlsalinewasadministeredintraperitoneallyonPTZgroup.Theratswerekilled.TostudytheinfluenceofMTonBcl-2andBaximmunoreactivityofneruononhippocampusbyusingimmunohistochemistrymethod.Results ThenumberandopticaldensityofBaxinhippocampusofPTZgroupratswashigherthanthatofcontrolgrouprats.ThenumberandopticaldensityofBaxinhippocampusofMTgroupratswaslowerthanthatofPTZgrouprats，washigherthanthatofcontrolgrouprats；ThenumberandopticaldensityofBcl-2inhippocampusofPTZgorupratswashigherthanthatofcontrolgrouprats；ThenumberandopticaldensityofBcl-2inhippocampusofMTgroupratswashigherthanthatofPTZgrouprats，washigherthanthatofcontrolgrouprats.Conclusion MelatonincanresistanttheapoptosisofhippocampusneruonbyincreasedtheexpressionofBcl-2anddecreasedtheexpressionofBax.      【Keywords】 melatonin；seizure；pentylenetetrazole；bax；bcl-2      难治性癫痫是神经系统的常见病，发病机制至
